Synchronous replication initiation of the two Vibrio cholerae chromosomes
The genome of Vibrio cholerae, the causative agent of cholera, is divided between two circular replicons. Genomic analysis has revealed that several other bacterial species have more than one chromosome. However, to date, the dynamics of chromosome replication in bacteria with more than one chromosome have not been investigated. As in V. cholerae chromosome II (1.07 Mb) is smaller than chromoso...

متن کاملNucleosomes influence multiple steps during replication initiation
Eukaryotic replication origin licensing, activation and timing are influenced by chromatin but a mechanistic understanding is lacking. Using reconstituted nucleosomal DNA replication assays, we assessed the impact of nucleosomes on replication initiation. To generate distinct nucleosomal landscapes, different chromatin-remodeling enzymes (CREs) were used to remodel nucleosomes on origin-DNA tem...
متن کاملPhosphorylated SIRT1 associates with replication origins to prevent excess replication initiation and preserve genomic stability
Chromatin structure affects DNA replication patterns, but the role of specific chromatin modifiers in regulating the replication process is yet unclear. We report that phosphorylation of the human SIRT1 deacetylase on Threonine 530 (T530-pSIRT1) modulates DNA synthesis. T530-pSIRT1 associates with replication origins and inhibits replication from a group of 'dormant' potential replication origi...
متن کاملMcm1 promotes replication initiation by binding specific elements at replication origins.
Minichromosome maintenance protein 1 (Mcm1) is required for efficient replication of autonomously replicating sequence (ARS)-containing plasmids in yeast cells. Reduced DNA binding activity in the Mcm1-1 mutant protein (P97L) results in selective initiation of a subset of replication origins and causes instability of ARS-containing plasmids. This plasmid instability in the mcm1-1 mutant can be ...
